In the world of rodents, the naked mole-rat stands out as an intriguing species that challenges our understanding of aging and cancer. Unlike other rodents in the Murinae group, which have a maximum lifespan of under five years, naked mole-rats have a lifespan that ranges between 10 to 30 years.

This longevity is a subject of ongoing research, with scientists believing that the gene responsible for the production of high-molecular-weight hyaluronic acid (HMW-HA) could be a key factor. A study published in 2023 found that mice with the naked mole-rat version of the HAS2 gene, which encodes for the enzyme responsible for producing HMW-HA, lived longer than those without it.

HMW-HA might explain naked mole-rats' apparent resistance to cancer. In the later mouse model study, mice with the naked mole-rat version of HAS2 were better protected against both spontaneous and purposely induced cancer. Interestingly, naked mole rat cells are more susceptible to becoming cancerous when HMW-HA isn't present.

The study, led by author Andrei Seluanov, also found that molecules that slow down hyaluronan degradation are being tested in pre-clinical trials. These findings could potentially be translated for use in humans to benefit human longevity and health in the future.

Naked mole-rats, however, still show one of the main molecular hallmarks of aging. Despite their impressive lifespan, they are not immune to the aging process. A study published in 2018 found that naked mole-rats do not follow the Gompertz law of mortality, a rule that generally applies to mammals. This suggests that naked mole-rats have a unique approach to aging that differs from other mammals.

The oldest reported naked mole-rat was 39 years old in 2021, a remarkable age for a rodent. However, the study did not provide information on whether Joe, the oldest reported naked mole-rat, is still alive.

While naked mole-rats may not have their looks going for them, they are certainly teaching scientists new things about aging and cancer. Their unique biology and longevity offer a wealth of information that could lead to breakthroughs in human health and longevity.
